Phospholipid Composition

Phospholipid composition refers to the types, proportions, and distribution of phospholipids within a biological membrane or lipid mixture. Because phospholipids contain hydrophilic heads and hydrophobic tails, they assemble into bilayers whose fluidity, thickness, curvature, and permeability depend on factors such as head-group chemistry, tail length, and degree of unsaturation. Cells regulate this composition to support membrane trafficking, signaling, compartmentalization, and interactions with proteins. Studying phospholipid composition helps explain how membranes adapt to environmental conditions and disease, while also informing research on liposomes, drug delivery systems, and synthetic membrane models.

Extraction and Analysis of Microbial Phospholipid Fatty Acids in Soils

Phospholipid fatty acids provide information about the structure of soil microbial communities. We present methods for extraction from soil samples with a single-phase chloroform mixture, fractionation of extracted lipids using solid phase extraction columns, and methanolysis to produce fatty acid methyl esters, which are analyzed by capillary gas chromatography.

Quantitative Flow Cytometry to Study Labeled Protein-Phospholipid Vesicle Interactions

In this video, we demonstrate the interaction between proteins and artificial phospholipid vesicles using quantitative flow cytometry. The binding of fluorescently-labeled proteins to fluorescently-labeled phospholipid vesicles increases the mean fluorescence intensity, and this increase is detected by a flow cytometer.
