Bioluminescence Tracking

Bioluminescence tracking is a noninvasive imaging method that follows living cells, tissues, or biological events by detecting light produced through a luciferase-luciferin reaction. In cancer research, scientists introduce luciferase genes into tumor cells or use reporter systems, then supply the required substrate; emitted photons are captured with sensitive cameras to estimate cell location and activity over time. This approach enables longitudinal monitoring of tumor growth, metastasis, treatment response, and cancer cell survival in animal models, often reducing the need to sacrifice animals at each time point. Its quantitative readouts support drug development and evaluation of disease mechanisms.

In fireflies, the luciferase enzyme converts a compound called luciferin into oxyluciferin, and produces light or “luminescence” as a result. This reaction requires energy derived from ATP in order to proceed, so researchers have exploited the luciferase-luciferin interaction to gauge ATP levels in cells. Improved understanding of pancreatic cancer biology is critically needed to enable the development of better therapeutic options to treat pancreatic cancer. To address this need, we demonstrate an orthotopic model of pancreatic cancer that permits non-invasive monitoring of cancer progression using in vivo bioluminescence imaging.

Herein we describe the methods to construct, visualize, and quantify the bioluminescent reactions of both firefly and renilla luciferase enzymes expressed in metastatic breast cancer cells during their growth and metastasis in vivo.
