Dimethyl sulfoxide (DMSO) is a polar aprotic organic solvent widely used in laboratory and medical research because it dissolves many polar and nonpolar compounds and readily penetrates biological membranes. In cancer research, DMSO commonly serves as a vehicle for preparing poorly water-soluble drugs, while its membrane permeability can influence cellular uptake and its concentration may affect cell viability, differentiation, or gene expression. Researchers therefore include matched vehicle controls and maintain carefully defined concentrations when testing anticancer compounds. DMSO is also used as a cryoprotectant for preserving cells, supporting cancer cell line storage and reproducible experimental workflows.

Generating differentiated cell types from human pluripotent stem cells (hPSCs) holds great therapeutic promise but remains challenging. PSCs often exhibit an inherent inability to differentiate even when stimulated with a proper set of signals. Described here is a simple tool to enhance multilineage differentiation across a variety of PSC lines.

Bacterial pyomelanin production results in increased resistance to oxidative stress and virulence. We report on techniques that can be used to determine inhibition of pyomelanin production and assay the resulting increase in sensitivity to oxidative stress in bacteria, as well as determine antibiotic minimum inhibitory concentration (MIC).
