Leukemic Cell Line C1498

Leukemic Cell Line C1498 is a murine myeloid leukemia cell line used as a laboratory model of blood cancer and tumor–immune interactions. Derived from a C57BL/6 mouse, C1498 cells can be expanded in culture and introduced into genetically compatible mice, where they establish leukemia and enable evaluation of disease progression. Researchers use this syngeneic system to study leukemia biology, antitumor immunity, immune-cell function, and experimental treatments in an immunocompetent setting. C1498 variants engineered to express defined antigens can also support investigations of antigen-specific T-cell responses, making the model valuable for cancer immunology and preclinical therapy research.

This manuscript provides a technical procedure that can be used to characterize C1498 cell cultures in vitro and the acute leukemia induced in mice after their injection. Phenotypic and functional analyses are performed using flow cytometry, immunofluorescence microscopy, cytochemistry and May-Grünwald Giemsa staining.

2D Co-culture of Leukemic Cells with BMSCs: A Method to Obtain Subpopulations of Leukemic Cells

2023

This video describes the technique for long term 2D co-culture of leukemia cells with bone marrow stromal cells to study their interaction with the bone marrow microenvironment. The 2D coculture model utilizes the acute lymphoblastic leukemia cells and the bone marrow stromal or osteoblasts that influence tumor cell phenotype. This study will support investigation of the mechanisms that underlie bone marrow supported survival of leukemic cells during chemotherapy exposure.

Leukemic Subpopulation Harvest: A Method for Spatial Separation of Leukemic Cell Subpopulations from 2D Co-culture

This video describes a method for spatial separation of three leukemic cell subpopulations: the suspended leukemic cells freely floating in the media, phase bright leukemic cells adhered to the surface of the mesenchymal stromal cell monolayer, and the phase dim leukemic cells that have migrated beneath the mesenchymal stromal cell monolayer from 2D co-culture.

Preparation of Subcellular Fractions from Leukemic Cells: A Method to Generate Cellular and Nuclear Fractions from Leukemic Cells
