Mouse Lung Cancer Models

Mouse lung cancer models are experimental systems used to reproduce key features of human lung tumors in a living organism, supporting cancer research from tumor initiation to treatment. Researchers generate these models through genetically engineered mutations, carcinogen exposure, transplantation of tumor cells, or implantation into lung tissue, with each approach capturing distinct aspects of tumor biology and the immune microenvironment. They help investigators study cancer development, progression, metastasis, and interactions between tumors and host tissues. Mouse models also enable preclinical evaluation of targeted therapies, chemotherapy, and immunotherapy, while providing a framework for identifying biomarkers and refining treatment strategies before clinical testing.

The goal of this paper is to describe simple methods that will greatly aid in the setup and analysis of mouse lungs with lung cancer or other pathologies. We present 3 protocols to simply and reliably carry out lung instillations, fixation, and lung volume measurements.

A Minimally Invasive Method for Generating a Syngeneic Orthotopic Mouse Model of Lung Cancer

A major challenge of developing new therapies for lung cancer includes modeling the tumor microenvironment as regional disease progresses to metastatic disease. Here, we describe a simple method of establishing an orthotopic syngeneic model of lung cancer that allows for in vivo tracking of disease over time.

We describe a method for the detection of tumor nodule development in the lungs of an adenocarcinoma mouse model using micro-computed tomography and its use for monitoring changes in nodule size over time and in response to treatment. The accuracy of the assessment was confirmed with end-point histological quantification.
