Prostate Carcinoma Pc3 Cells

Prostate carcinoma PC3 cells are a human prostate cancer cell line derived from a bone metastasis and widely used to model advanced, androgen-independent disease. Their growth does not depend on functional androgen receptor signaling, allowing researchers to examine cancer behaviors that persist despite hormone-directed treatment. In culture, PC3 cells support assays of proliferation, survival, migration, invasion, tumor signaling, and response to anticancer compounds. These properties make the line valuable for studying metastatic progression and therapeutic resistance, while results must be interpreted alongside other prostate cancer models because a single cell line cannot represent the full biological diversity of the disease.

We describe the manufacture of compressed hormone pellets, as well as subcutaneous surgical implantation into mice. This strategy can be combined with the growth of cell and tissue xenografts under the renal capsule of athymic nude mice to evaluate hormonal carcinogenesis and regulation of benign prostate growth.
