Alpha Fractions

Alpha fractions are the proportions of a chemical substance present as each protonation state at equilibrium, providing a quantitative description of acid–base speciation. Their values depend on the species’ dissociation constants and the solution pH: for a polyprotic acid, successive deprotonation steps redistribute the total analytical concentration among forms such as H₂A, HA⁻, and A²⁻. Chemists use alpha fractions to predict the dominant species across a pH range, interpret titration curves, design buffer systems, and assess how protonation influences solubility, reactivity, and metal-ion complexation. These calculations connect equilibrium theory with practical control of chemical behavior in laboratory and industrial systems.

Here, we present a protocol for the isolation of increasingly insoluble/aggregated alpha-synuclein (α-syn) from post-mortem human brain tissue. Through the utilization of buffers with increasing detergent strength and high-speed ultracentrifugation techniques, the variable properties of α-syn aggregation in diseased and non-diseased tissue can be examined.

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain.
