Blocking Peptide

A blocking peptide is a short peptide designed to bind a target antibody or molecular recognition site and prevent another molecule from binding, making it a useful tool for testing molecular specificity. In a typical assay, the peptide is added before or alongside the sample; if it occupies the antibody’s antigen-binding site through complementary interactions, such as hydrogen bonding and electrostatic contacts, the target signal should decrease or disappear. In chemistry and biochemical research, this competition control helps determine whether an antibody signal reflects the intended epitope rather than nonspecific binding, supporting assay validation, protein characterization, and more reliable interpretation of experimental results.

Development of a dual-functional conjugate of antigenic peptide and Fc-III mimetics (DCAF) is novel for the elimination of harmful antibodies. Here, we describe a detailed protocol for the synthesis of DCAF1 molecule, which can selectively block 4G2 antibody to eliminate antibody dependent enhancement effect during Dengue virus infection.

Peptide Purification: An RP-HPLC-based Technique to Extract Peptides from Digested Protein Lysates

In this video, we demonstrate reversed-phase high-performance liquid chromatography to purify peptides from a digested protein lysate mixture. These purified peptides are then lyophilized and stored for downstream applications. Analysis of peptides can help understand the mechanics involved in cellular signaling and cancer.

A Peptide Microarray Technology for Specificity Profiling of Antibodies

2025

This video demonstrates a technique that applies peptide microarray technology to profile the specificity of antibodies recognizing post-translational modifications (PTMs) on histone peptides. A microarray slide containing immobilized fluorophore-conjugated histone peptides with known PTM combinations is treated with target PTM-specific antibodies. This process helps identify the antibody's specificity for the target and its potential cross-reactivity with non-target PTMs.
