Cytochrome P450 Enzymes

Cytochrome P450 enzymes are heme-containing proteins that catalyze the oxidation of many drugs, hormones, and other chemicals, making them central to clinical pharmacology. Using electrons supplied by NADPH through a reductase partner, these enzymes activate molecular oxygen and insert one oxygen atom into a substrate while reducing the other to water. Their activity influences drug clearance, therapeutic effectiveness, and toxicity, while genetic variation, liver disease, and enzyme inhibition or induction can alter patient responses. Understanding P450-mediated metabolism supports dose selection, prediction of drug interactions, and development of safer, more individualized treatments.

Activation of Apoptosis by Cytoplasmic Microinjection of Cytochrome c

In this protocol, we describe the direct cytoplasmic microinjection of cytochrome c protein into fibroblasts and primary sympathetic neurons. This technique allows for the introduction of cytochrome c protein into the cytoplasm of cells and mimics the release of cytochrome c from mitochondria, which occurs during apoptosis.

This procedure describes how to encapsulate cytochrome c (cyt. c) in silica (SiO2) sol-gels, process these gels to form bioaerogels, and use these bioaerogels to rapidly recognize nitric oxide (NO) through a gas-phase reaction. Biological Catalysts All living organisms continuously perform numerous biochemical reactions to sustain their presence. Most of these reactions require an input of energy to start, which is called the activation energy. Catalysts are chemicals that lower the activation energy. Even though catalysts facilitate a chemical reaction, they are not consumed by it. This means a catalyst can facilitate a specific chemical reaction over and over in succession, increasing the rate of that reaction.
