In Vitro In Vivo Correlation

In Vitro In Vivo Correlation (IVIVC) is a predictive relationship between a drug’s in vitro dissolution or release behavior and its in vivo absorption, helping connect laboratory testing with clinical performance. It works by comparing dissolution profiles from dosage forms with pharmacokinetic measures such as plasma drug concentration, exposure, or absorption rate, often through mathematical modeling and deconvolution. In clinical development, IVIVC can support formulation optimization, bioequivalence assessment, dissolution specifications, and decisions about the need for additional in vivo studies. By improving prediction of drug performance, it can streamline development while reducing reliance on extensive clinical testing.
