Respiratory Diagnostics

Respiratory diagnostics encompasses clinical tests used to evaluate the structure and function of the airways and lungs, helping identify causes of breathing problems. These methods measure physiological signals such as airflow, lung volume, gas exchange, and blood oxygen saturation through procedures including spirometry, imaging, and pulse oximetry. Clinicians use the results to detect, classify, and monitor conditions such as asthma, chronic obstructive pulmonary disease, infections, and respiratory failure. By combining patient history with objective measurements, respiratory diagnostics supports timely treatment decisions, tracks disease progression, and helps assess responses to therapy.

Here, we present a protocol for the development and validation of a quantitative PCR method used for the detection and quantification of EHV-2 DNA in equine respiratory fluids. The EHV-2 qRT-PCR validation protocol involves a three-part procedure: development, characterization of qRT-PCR assay alone, and characterization of the whole analytical method.

A rapidly-deployable, off-grid laboratory has been designed and built for remote, resource-constrained global settings. The features and critical aspects of the logistically-enhanced, expandable, multifunctional laboratory modules are explored. A checklist for a basic laboratory workflow and a protocol for a respiratory viral diagnostic test are developed and presented.

Diagnostic ultrasound imaging has proven to be effective in diagnosing various respiratory diseases in human and animal subjects. We demonstrate a comprehensive ultrasound protocol utilized by Dr. Zuo's lab to analyze diaphragm kinetics specifically in mouse models. This is also a non-invasive research technique which can provide quantitative information on mouse respiratory muscle function.
