Microwell Plate Technology

Microwell plate technology uses arrays of small, uniformly sized wells to organize cells, embryos, or other biological samples under controlled laboratory conditions. By confining specimens within defined microwells, the method supports consistent cell aggregation, limits unwanted movement, and enables parallel observation or treatment across many experimental units. In developmental biology, microwell plates help generate and study embryoid bodies, organoids, and early tissue structures while controlling factors such as cell number, spatial arrangement, and exposure to signaling molecules. This standardization improves reproducibility and facilitates high-throughput analysis of cell differentiation, morphogenesis, developmental timing, and responses to genetic or chemical perturbations.

Conventional methods to initiate suspension aggregate based cardiac differentiation of human pluripotent stems cells (hPSCs) are plagued with culture heterogeneity with respect to aggregate size and shape. Here, we describe a robust method for cardiac differentiation employing microwells to generate size-controlled hPSC aggregates cultured under cardiac-promoting conditions.

A Paired Bead and Magnet Array for Molding Microwells with Variable Concave Geometries

This manuscript introduces a robust method of fabricating concave microwells without the need for complex high-cost facilities. Using magnetic force, steel beads, and a through-hole array, several hundred microwells were formed in a 3 cm x 3 cm polydimethylsiloxane (PDMS) substrate.
