Gfp Reporter Assay

A GFP reporter assay is a molecular technique that uses green fluorescent protein to visualize gene expression, protein localization, or cellular activity in living specimens. Researchers place the GFP coding sequence under the control of a selected promoter or regulatory element; when that element is active, cells produce GFP, which emits green fluorescence after excitation with blue or ultraviolet light. In developmental biology, this assay reveals when and where genes are activated during embryogenesis, tissue formation, and cell differentiation. Time-resolved fluorescence imaging can track developmental patterns and help connect gene regulation with cell behavior and tissue organization.

Source: Chen, Y., et al., Measurement of Specific Mycobacterial Mistranslation Rates with Gain-of-function Reporter Systems. J. Vis. Exp. (2019)This video demonstrates a method to quantify mistranslation in Mycobacterium smegmatis using an inducible GFP and secreted mutant luciferase reporter system. It shows how luminescence resulting from mistranslation is measured and compared to GFP fluorescence to evaluate the impact of a test molecule on translational accuracy.
