Morpholino-mediated Knockdown

Morpholino-mediated knockdown is a gene-function analysis technique that reduces expression of a target gene using synthetic antisense oligonucleotides, making it useful for testing gene roles in developing organisms. Morpholinos bind complementary RNA sequences through base pairing but resist nuclease degradation; depending on their target site, they sterically block translation initiation or alter pre-mRNA splicing rather than trigger RNA cleavage. In developmental biology, researchers introduce morpholinos into embryos and examine changes in cell fate, tissue formation, or organ development, often comparing dose responses with mismatch controls and rescue experiments. This approach can reveal developmental requirements for specific genes, although incomplete knockdown and off-target effects can complicate interpretation.

Lentiviral-mediated Knockdown During Ex Vivo Erythropoiesis of Human Hematopoietic Stem Cells

An ex vivo protocol to generate mature human red blood cells from hematopoietic stem/progenitors is described. Additionally we describe an efficient lentiviral-delivery method to knockdown the transcription factor TAL1 in primary erythroid cells. The efficiency of lentivirus mediated gene delivery is demonstrated using GFP expressing viruses.

This protocol aims to illustrate the techniques needed to introduce stably transmitted DNA and transient double stranded RNA into the necromenic nematode Pristionchus pacificus for studies in evolutionary, developmental, and behavioral biology.
