Optic Axon Arborization

Optic axon arborization is the developmental process by which retinal ganglion cell axons branch and form terminal arbors within specific visual targets, establishing precise neural connections for vision. After extending along defined pathways, axons respond to molecular guidance cues and neural activity that regulate target recognition, branching, synapse formation, and refinement. Studying this process in developmental biology reveals how visual circuits achieve spatial and functional organization, and how disrupted axon targeting or arbor structure can impair visual processing. Research on optic axon arborization also provides broader insight into neural wiring, activity-dependent circuit refinement, and mechanisms of developmental connectivity.

Here, we describe a protocol for visualizing motor neuron projection and axon arborization in transgenic Hb9::GFP mouse embryos. After immunostaining for motor neurons, we used light sheet fluorescence microscopy to image embryos for subsequent quantitative analysis. This protocol is applicable to other neuron navigation processes in the central nervous system.
