Structural Electrophysiological Maturation

Structural electrophysiological maturation is the developmental process through which excitable cells acquire the morphology, membrane properties, and signaling capacity required for coordinated function. It occurs as cells remodel their shape and connectivity while changing ion-channel expression, membrane resistance, capacitance, and action-potential dynamics, linking structural development with electrical activity. In developmental biology, studying these changes helps characterize the progression of neurons, cardiomyocytes, and other electrically active cells from immature states to functional phenotypes. Measurements such as microscopy, patch-clamp recording, and extracellular electrophysiology can reveal maturation deficits, support validation of stem-cell-derived models, and advance research on congenital disorders, tissue engineering, and regenerative therapies.

Here, we describe the preparation of viable ventricular slices from adult mice and their use for sharp electrode action potential recordings. These multicellular preparations provide a preserved in vivo like tissue structure, which makes them a valuable model for electrophysiological and pharmacological studies in vitro.
