Dual Beam Fib

Dual beam FIB (focused ion beam) is an engineering characterization and fabrication system that combines a focused ion beam with a scanning electron microscope (SEM), enabling precise material removal alongside high-resolution imaging. The ion beam sputters atoms from a selected region for milling, cross-sectioning, or site-specific preparation, while the electron beam images surface structure and guides process control; some systems also support ion- or electron-induced deposition. This coordinated workflow supports semiconductor failure analysis, microscale machining, circuit editing, and preparation of electron-transparent specimens for transmission electron microscopy, linking three-dimensional structure to material performance.

A description of how to calibrate Förster Resonance Energy Transfer integrated biological sensors (FIBS) for in situ metabolic profiling is presented. The FIBS can be used to measure intracellular levels of metabolites noninvasively aiding in the development of metabolic models and high throughput screening of bioprocess conditions.

Workflow Using a Cryogenic Coincident Fluorescence, Electron, and Ion Beam Microscope for Targeted Milling of Cells

This workflow enables lamella production targeting fluorescently labeled biological structures that are small (<1 μm in axial extent) and rare (1 copy per cell) using a cryogenic tri-coincident imaging platform. This platform integrates fluorescence microscopy, focused ion beam milling, and scanning electron microscopy at a single focal position and enables simultaneous fluorescence microscopy while milling.
