Eastern Painted Turtle

The Eastern Painted Turtle (Chrysemys picta picta) is a freshwater turtle native to eastern North America and an important component of wetland ecosystems. As an ectotherm, it regulates body temperature by basking in sunlight and retreating into water, while aquatic habitats provide food, shelter, and sites for reproduction; during winter, it can remain inactive in submerged, low-oxygen conditions. Studying this turtle helps researchers assess wetland health, understand responses to habitat loss and water-quality changes, and evaluate how temperature influences behavior, growth, and sex determination. Its conservation therefore links freshwater management with broader environmental monitoring.

2D and 3D Chromosome Painting in Malaria Mosquitoes

Chromosome painting is a useful method for studying organization of the cell nucleus and evolution of the karyotype. Here, we demonstrate an approach to isolate and amplify specific regions of interest from single polytene chromosomes that are subsequently used for two- and three-dimensional fluorescent in situ hybridization (FISH).

Ocular Kinematics Measured by In Vitro Stimulation of the Cranial Nerves in the Turtle

This protocol describes how to use an in vitro isolated turtle head preparation to measure the kinematics of their eye movements. After removal of the brain from the cranium, cranial nerves can be stimulated with currents to quantify rotations of the eye and changes in pupil sizes.
