Egfp Expression Analysis

EGFP expression analysis is the measurement of enhanced green fluorescent protein production as an indicator of gene activity in cells or organisms. The method relies on an EGFP reporter gene whose protein product fluoresces green when excited with blue or ultraviolet light, allowing expression levels to be detected through fluorescence microscopy, plate readers, or flow cytometry. In environmental research, EGFP can reveal how microorganisms or cultured cells respond to pollutants, nutrients, temperature, or other conditions by linking fluorescence intensity to regulatory or physiological changes. This approach supports studies of environmental stress, microbial function, gene regulation, and the development of fluorescent biosensors for monitoring ecosystems and contaminants.

Development of Leishmania Species Strains with Constitutive Expression of eGFP

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.

In Parkinson's Disease (PD), Substantia Nigra (SNc) dopaminergic neurons degenerate, leading to motor dysfunction. Here we report a protocol for culturing ventral midbrain neurons from a mouse expressing eGFP driven by a Tyrosine Hydroxylase (TH) promoter sequence, harvesting individual fluorescent neurons from the cultures, and measuring their transcriptome using RNA-seq.

Expression Analysis of Mammalian Linker-histone Subtypes

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.
