Embryo Grafting

Embryo grafting is a technique in which an excised plant embryo is transferred onto a compatible rootstock so it can continue developing, making it useful for studying inheritance and rescuing otherwise nonviable hybrids. Under sterile conditions, the immature embryo is carefully isolated and placed in contact with rootstock tissue, which supplies water, minerals, and growth support as the graft establishes. In plant genetics, the method helps recover hybrids from wide crosses, overcome embryo abortion or seed dormancy, and maintain valuable genotypes that cannot mature normally in intact seeds. It also supports research on embryo development, compatibility, and the genetic control of early plant growth.

Grafting of Beads into Developing Chicken Embryo Limbs to Identify Signal Transduction Pathways Affecting Gene Expression

By grafting beads soaked in growth factors or specific inhibitors of signaling pathways into developing embryos it is possible to directly test their effects in vivo. In this protocol beads are grafted into the limb bud to determine the effects of these molecules on gene expression and signal transduction.

Here we report dual labeling of neural crest cells and blood vessels using chickGFP neural tube intraspecies grafting combined with intra-vascular DiI injection. This experimental technique allows us to simultaneously visualize and study development of the NCC-derived (enteric) nervous system and the vascular system, during organogenesis.

Neural induction is the first step in the formation of the brain. It is a mechanism by which Hensen's node (organizer), instructs adjacent tissue to adopt a neural fate, i.e. to give rise to the nervous system. This video demonstrates an assay for neural induction in chick embryo.

Mouse Models for Graft Arteriosclerosis

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.
