Lentiviral Transgenesis

Lentiviral transgenesis is a genetic engineering method that introduces a transgene into a cell’s genome using a modified lentiviral vector, enabling stable inheritance and expression of the added sequence. After entering target cells, the vector’s RNA genome is reverse-transcribed into DNA and integrated into host chromosomes by the viral integrase, while pathogenic viral genes are removed from the delivery system. In genetics, this approach supports gene-function studies, creation of transgenic cell lines and model organisms, and investigation of disease-associated variants. Its ability to transduce both dividing and nondividing cells makes it valuable for long-term experiments in research and biotechnology.

This protocol aims to illustrate the techniques needed to introduce stably transmitted DNA and transient double stranded RNA into the necromenic nematode Pristionchus pacificus for studies in evolutionary, developmental, and behavioral biology.
