Mt-atp6 Variant

An MT-ATP6 variant is a change in the mitochondrial gene MT-ATP6, which encodes a subunit of ATP synthase and can affect cellular energy production. Because mitochondrial DNA is inherited differently from most nuclear genes and may be present in mixed normal and altered populations, a condition known as heteroplasmy, the effects of a variant can vary among tissues and individuals. Genetic analysis of MT-ATP6 variants supports research on mitochondrial disorders, oxidative phosphorylation, and genotype-phenotype relationships, while laboratory and clinical interpretation help distinguish potentially pathogenic changes from benign variation and clarify mechanisms of mitochondrial disease.

Histone Variants at the Centromere

2020

Histone variants are the histone proteins with structural and sequence variations. These variants may be regarded as “mutant” forms that replace their canonical histone counterparts in the nucleosomes. Specific post-translational modifications on the histone variants enable further chromatin complexity and regulate tissue-specific gene expression. The most common histone variants are from histone H2A, H2B, and linker histone H1 families. However, several variants of histone H3 variants are also...

2012

Pooled DNA sequencing is a fast and cost-effective strategy to detect rare variants associated with complex phenotypes in large cohorts. Here we describe the computational analysis of pooled, next-generation sequencing of 32 cancer-related genes using the SPLINTER software package. This method is scalable, and applicable to any phenotype of interest.

Generation of Influenza Virus Escape Variants against Neutralizing Antibodies

2025

This video demonstrates a technique for generating escape variants of neutralizing influenza virus monoclonal antibodies. Antibody-neutralized virus suspension is injected into embryonated chicken eggs to propagate escape variants selectively. This results in a virus population predominantly of escape variants, confirmed via a Hemagglutination assay.

We describe a method by which we identify critical residues required for the binding of human or murine monoclonal antibodies that target the viral hemagglutinin of influenza A viruses. The protocol can be adapted to other virus surface glycoproteins and their corresponding neutralizing antibodies.

Assessing the Antimicrobial Activity of Nisin Variants Using an Agar Well Diffusion Assay

Source: Nickling, J. H., et.al. Antimicrobial Peptides Produced by Selective Pressure Incorporation of Non-canonical Amino Acids. J. Vis. Exp. (2018)This video demonstrates an agar well diffusion assay used to evaluate the antimicrobial activity of nisin variants. It shows how Lactococcus lactis expressing the membrane-bound peptidase NisP activates nisin precursors produced in E. coli, resulting in growth inhibition halos. The size of these halos reflects the bioactivity of each nisin variant.
