Antibody Localization

Antibody localization is the determination of where antibodies or their target antigens are distributed within cells, tissues, or infected specimens, providing spatial context for immune responses. In immunology, labeled primary or secondary antibodies bind specific epitopes, and imaging methods such as immunofluorescence or immunohistochemistry reveal these binding sites through fluorescence or enzymatic color development. Researchers use antibody localization to identify pathogen components, distinguish intracellular from extracellular targets, and map immune-cell or antigen distribution in tissue sections. These observations help clarify infection biology, evaluate host responses, and support diagnostic and experimental studies of immune mechanisms.

A Technique to Assess the In Vivo Localization of Tumor-Specific Antibodies

2025

This video demonstrates a technique to study the in vivo localization of tumor-specific antibodies in a mouse tumor xenograft model. Upon subcutaneously injecting cancer cells into mice for tumor formation, fluorescently-labeled antibodies are injected that bind to tumor-specific receptors, enabling in vivo assessment of antibody localization via imaging.

Single-Molecule Localization Microscopy of Membrane Proteins using Single-Antibody Labeling

2026

This protocol describes single-antibody labeling (SAL) to resolve the nanoscale spatial organization of plasma membrane proteins. By leveraging cumulative antibody-epitope interactions at the single-molecule level, membrane SAL (mSAL) maps local epitope distributions while simultaneously capturing antibody binding behavior in the native cellular environment.

In Vivo Fluorescence Imaging to Localize Antibodies in a Mouse Tumor Xenograft Model

2025

The video demonstrates a fluorescence imaging method for in vivo antibody localization in a mouse tumor xenograft model. It involves the injection of tumor cells in a mouse to generate solid xenografts beneath the skin. Subsequent injection of fluorescent antibodies confirms specific localization through enhanced fluorescence signals within the tumor xenograft.

An In Vivo Immunofluorescence Localization Method to Study Antibody Biodistribution

2025

This video demonstrates an in vivo immunofluorescence localization (IVIL) technique to study the biodistribution of tumor-targeting antibodies in a murine model of breast cancer. Upon the intravenous injection of a tumor-specific antibody in the mouse, the tumor is surgically isolated, and the localization of the antibody within the tumor tissue is confirmed via ex vivo immunostaining.
