Biofilm Visualization

Biofilm visualization comprises methods for observing the organization, development, and activity of microbial communities enclosed in a self-produced extracellular matrix. These approaches use fluorescent or chemical stains, microscopy, and sometimes three-dimensional imaging to distinguish cells, matrix components, viability, and spatial structure within surface-associated communities. In immunology and infection research, visualization reveals how biofilms attach to tissues or medical devices, form protective barriers, and interact with immune cells and antimicrobial agents. The resulting structural and functional information helps explain persistent infections, assess treatment responses, and support the development of strategies that disrupt biofilm formation or improve immune and therapeutic access.

Mycobacterium tuberculosis forms drug tolerant biofilms when cultured in certain conditions. Here we describe methods for culturing M. tuberculosis biofilms and determining the frequency of drug tolerant persisters. These protocols will be useful for further studies into the mechanisms of drug tolerance in M. tuberculosis.
