Borrelia Species

Borrelia species are spiral-shaped bacteria in the family Spirochaetaceae, including agents of Lyme disease and relapsing fever that shape major questions in infection and immune defense. They move through tissues using periplasmic flagella and persist in hosts by altering surface antigens, helping them evade antibody-mediated clearance while interacting with tick vectors and vertebrate reservoirs. In immunology and infection research, Borrelia species support studies of pathogen transmission, host inflammation, antigenic variation, and protective immunity. Species-level identification through culture, molecular assays, or sequencing improves diagnosis and epidemiological surveillance, while mechanistic insight informs vaccine development and treatment strategies.

Source: Eggers, C. H. Phage-Mediated Genetic Manipulation of the Lyme Disease Spirochete Borrelia burgdorferi. J. Vis. Exp. (2022).This video demonstrates the isolation of borrelial phage particles from a Borrelia burgdorferi suspension using polyethylene glycol-mediated phase separation. The sample is centrifuged, treated with a salt solution, and a molecular crowding agent to precipitate phages. Chloroform extraction is done to remove debris and concentrate phage particles. The purified phage...

Source: Eggers, C. H. Phage-Mediated Genetic Manipulation of the Lyme Disease Spirochete Borrelia burgdorferi. J. Vis. Exp. (2022)This video demonstrates the preparation of a genetically modified Borrelia burgdorferi culture engineered with dormant bacteriophage DNA carrying an antibiotic resistance gene. Following antibiotic selection and incubation, a chemical inducer triggers DNA damage, leading to excision, replication, and assembly of new virions containing the resistance gene. After a...

Simulating Colonization and Extinction on Islands ExpandForm groups of three. One member of the group will simulate colonization by throwing balls into the groups of cups representing the islands. Another will simulate extinction by rolling dice to randomly remove balls from the cups. The third group member will record the data. Take time to familiarize yourself with the four island stations. NOTE: There are two large islands with 12 cups and two small islands with 6 cups. Two of the stations...

Species Distribution and Biogeography - Concepts

Species Distribution Biogeography is the study of species distribution across geographic space and the processes that shape these distributions. This discipline is based on the assumption that each species within a location must have immigrated from another geographic area or evolved from a local species. Within each habitat, a variety of biotic and abiotic factors act on individual species to shape birth, death, immigration, and emigration rates. To avoid local extinction, the combined birth...

In vitro culture is a direct detection method for the presence of living bacteria. This protocol describes methods for the culture of diverse Borrelia spirochetes, including those of the Borrelia burgdorferi sensu lato complex, relapsing fever Borrelia species, and Borrelia miyamotoi. These species are fastidious and slow growing but can be cultured.
