High-throughput Testing

High-throughput testing is a method for analyzing large numbers of biological samples or experimental conditions rapidly and systematically, enabling efficient measurement of complex immune and infectious processes. It combines miniaturized assays, automation, robotics, standardized workflows, and computational data analysis to process many samples in parallel while maintaining consistency and quality control. In immunology and infection research, high-throughput testing can compare antibody or cytokine responses, screen host-pathogen interactions, evaluate antimicrobial or antiviral compounds, and identify diagnostic biomarkers. These approaches increase experimental scale, reduce processing time and sample use, and generate data that support quantitative comparisons, mechanistic studies, and the development of new therapeutics or vaccines.

2016

A high throughput, real-time assay was developed to simultaneously identify (1) eukaryotic cell-penetrant antimicrobials targeting an intracellular bacterial pathogen, and (2) assess eukaryotic cell cytotoxicity. A variation on the same technology was thereafter combined with digital dispensing technology to enable facile, high-resolution, dose-response, and two- and three-dimensional synergy studies.

This protocol details high-throughput crystallization screening, ranging from the 1,536 microassay plate preparation to the end of a 6 week experimental time window. Details are included about the sample setup, the imaging obtained, and how users can perform analyses using an artificial intelligence-enabled graphical user interface to quickly and efficiently identify macromolecular crystallization conditions.

A Novel High-Throughput Ex Vivo Ovine Skin Wound Model for Testing Emerging Antibiotics

2022

The protocol describes a step-by-step method to set up an ex vivo ovine wounded skin model infected with Staphylococcus aureus. This high-throughput model better simulates infections in vivo compared with conventional microbiology techniques and presents researchers with a physiologically relevant platform to test the efficacy of emerging antimicrobials.

Anxiety Testing

2023

Anxiety is a commonly observed behavioral disorder that stems from fear. It is described as increased restlessness, or unpleasant feelings of fear over anticipated events. Experimenters often use rodent models to better understand anxiety disorders in humans. They use different paradigms, like exposing rodents to bright spaces or loud sounds, which are known to induce fear. These tests combined with other interventions such as surgery or drug-administration may assist researchers in pinpointing...
