Innate T Cells

Innate T cells are T lymphocytes that combine features of adaptive T cells with the rapid, broadly reactive behavior of innate immune cells, making them important early responders to infection. Unlike conventional T cells, subsets such as natural killer T, mucosal-associated invariant T, and γδ T cells use relatively restricted receptors to recognize lipid, metabolite, or stress-associated signals presented by nonclassical molecules or expressed by infected and damaged cells. Upon activation, they rapidly release cytokines and deploy cytotoxic mechanisms that shape inflammation and support other immune cells. Studying innate T cells helps clarify host defense, immunopathology, vaccine responses, and their potential for targeted immunotherapies.

Unlike cell-free HIV-1 particles, infected CD4+ T cells are effectively sensed by plasmacytoid dendritic cells (pDCs). This manuscript describes a method where peripheral blood mononuclear cells (PBMCs) or isolated pDCs are co-cultured with HIV-1 infected T cells to evaluate innate sensing by pDCs as assessed by release of type-I IFN.
