Hibit Bioreporter

Hibit Bioreporter is a luminescent protein-reporting system used to monitor biological events in living cells, including interactions between pathogens and host immune pathways. It uses a small HiBiT peptide genetically fused to a protein of interest; when the complementary LgBiT component and substrate are present, the two fragments assemble into an active luciferase and produce light in proportion to the tagged protein’s presence or activity. In immunology and infection research, this approach supports sensitive, quantitative measurement of protein abundance, trafficking, degradation, and host-pathogen interactions. Its compact tag can reduce disruption of target proteins while enabling real-time assays and higher-throughput analysis.

The Luciferase Assay: A High-Throughput Assay to Measure Luminescent Signals Using Engineered Nanoluciferase System-Based Bioreporters

The video demonstrates the luciferase assay used to quantify luciferase-tagged proteins. The assay uses an engineered luciferase enzyme composed of a large fragment that binds with the small fragment, which is conjugated with a protein of interest. The bound subunits form an active enzyme that, in the presence of the substrate, releases a luminescent signal. This signal is then quantified using a luminometer.
