Leukocyte Enrichment

Leukocyte enrichment is a laboratory process that increases the proportion of white blood cells in a blood or tissue sample, enabling more focused analysis of immune responses. It typically separates leukocytes from red blood cells, plasma, and other components using differences in density, size, or cell-surface markers, with methods such as density-gradient centrifugation or antibody-based magnetic separation. In immunology and infection research, enriched leukocyte preparations support flow cytometry, cell culture, immune profiling, and pathogen-response studies. By improving cell recovery and reducing sample complexity, the technique helps researchers characterize immune-cell populations and investigate inflammation, host defense, and disease mechanisms.

Fast and Specific Assessment of the Halogenating Peroxidase Activity in Leukocyte-enriched Blood Samples

This protocol describes the quick enrichment of leukocytes from small blood samples for a subsequent specific determination of the halogenating peroxidase activity within the cells. Isolation of Brain-infiltrating Leukocytes

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.

Marginating-hepatic leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe a method for selective harvesting of this specific hepatic cell population, through forced perfusion of the liver of rats or mice. Marginating-hepatic leukocytes seem critical in determining susceptibility to hepatic-related diseases and metastases.
