Lymphocyte Culture

Lymphocyte culture is a laboratory method for maintaining and studying lymphocytes outside the body under controlled conditions, making it important for investigating immune function and infection. In a typical culture, isolated lymphocytes are placed in nutrient-rich medium and may receive antigenic, mitogenic, or cytokine stimulation that promotes cell survival, activation, proliferation, and differentiation. Researchers use cultured lymphocytes to examine immune responses to pathogens, measure cytokine production, evaluate cellular interactions, and test therapeutic or diagnostic approaches. The technique supports studies of adaptive immunity, host-pathogen interactions, immunodeficiency, inflammation, and vaccine responses while providing a controlled system for analyzing cellular mechanisms.

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.
