Murine Bal Fluid

Murine BAL fluid, or bronchoalveolar lavage fluid, is a sample recovered from the lower airways of mice to assess the pulmonary environment during health, inflammation, or infection. During lavage, sterile fluid is introduced through the trachea and withdrawn, collecting airway cells, soluble proteins, microbial components, and other materials from the alveolar space. Researchers analyze these constituents through cell counts, cytology, protein or cytokine assays, and pathogen detection to characterize immune-cell recruitment, barrier injury, and inflammatory responses. In immunology and infection research, murine BAL fluid helps evaluate disease mechanisms, compare experimental treatments, and monitor how respiratory tissues respond over time.

We describe a research technique for fiberoptic bronchoscopy and bronchoalveolar lavage using low pressure suction. The technique is used to harvest immune cells from the lung bronchoalveolar surfaces. Local anesthetic and mild conscious sedation (midazolam) is used. Subjects tolerate the procedure well and experience minimal side effects.

Repeated measurements of rodent respiratory physiology and sampling of airway inflammatory cells are desirable, but generally not feasible. Here we describe a repeatable method for orally intubating mice that permits repeated measurements of airway hyperreactivity and sampling of airway inflammatory cells.

Murine Nasal Lavage Fluid Collection without Blood Contamination

The present protocol describes a novel method for avoiding blood contamination in murine nasal lavage fluid (NLF). Since this method ensures the NLF collection without blood contamination, it allows for more accurate detections of immunological components and various respiratory pathogens.

The health status of the lung is reflected by the type and number of immune cells that are present in the bronchioles of the lung. We describe a bronchoalveolar lavage technique that allows the isolation and study of nonadherent cells and soluble factors from the lower respiratory tract of mice.
