Plant Virus Identification

Plant virus identification is the process of detecting and distinguishing viruses that infect plants, a critical step in diagnosing disease, protecting crops, and tracing infection. It combines symptom assessment with laboratory methods such as antibody-based assays, including enzyme-linked immunosorbent assays, and nucleic acid amplification, which detect viral proteins or genetic sequences in plant tissue. In immunological testing, antibodies bind specific epitopes on viral particles or coat proteins, producing a measurable signal that indicates infection. Accurate identification supports disease management, surveillance, pathogen characterization, and development of resistant cultivars, while helping researchers understand host-pathogen interactions and immune responses in plants.

Improving our understanding of and our ability to manage many of the insect-transmitted plant viruses requires the use of the vector. Insect transmission of plant viruses is a tritrophic interaction, and as such requires the manipulation of insects, virus, and plant. Vectors must be reared in large numbers and manipulated in such a way as to insure high rates of transmission to test plants. The basics of rearing and manipulating the whitefly, Bemisa tabaci (Hemiptera: Aleyrodidae), a vector of...

Source: Zhao, J., et al. Potato Virus X-Based microRNA Silencing (VbMS) In Potato. J. Vis. Exp. (2020).This video demonstrates the use of Agrobacterium tumefaciens to deliver a Potato Virus X (PVX)-based construct into in vitro-grown potato plants for virus-based microRNA silencing (VbMS). The method enables systemic suppression of a target microRNA, allowing researchers to study its role in plant development through observable phenotypic changes.
