Platelet-leukocyte Interactions

Platelet-leukocyte interactions are coordinated contacts between circulating platelets and immune cells that link hemostasis with inflammation and host defense. Following vascular injury or infection, platelet activation exposes P-selectin and other adhesion molecules, enabling binding to leukocyte receptors, promoting leukocyte recruitment, and stimulating the release of cytokines, chemokines, and antimicrobial factors. These interactions influence vascular inflammation, thrombosis, pathogen containment, and tissue injury, making them important in immunology and infection research. Studying platelet-leukocyte aggregates can help clarify disease mechanisms and support the development of biomarkers or therapies for sepsis, inflammatory disorders, and thromboinflammatory conditions.

Blood leukocytes and platelets can be used as a marker of overall bioenergetic health of an individual and so have the potential to monitor pathological processes and the impact of treatments. Here we describe a method to isolate and measure mitochondrial function and the oxidative burst in these cells.

A Laminar Flow-Based Assay to Study the Interactions Between Leukocytes and Endothelial Cells

2025

This video demonstrates a laminar flow-based assay to characterize the interaction of leukocytes and cellular partners such as endothelial cells. This method provides dynamic insights into adhesion, deadhesion, and transmigration of leukocytes when perfused over an intact and confluent layer of vascular cells.

Real-time Imaging of Heterotypic Platelet-neutrophil Interactions on the Activated Endothelium During Vascular Inflammation and Thrombus Formation in Live Mice

Here we report an experimental technique of fluorescence intravital microscopy to visualize heterotypic platelet-neutrophil interactions on the activated endothelium during vascular inflammation and thrombus formation in live mice. This microscopic technology will be valuable to study the molecular mechanism of vascular disease and to test pharmacologic agents under pathophysiological conditions.
