Viral Antigen Plasmids

Viral antigen plasmids are circular DNA molecules engineered to carry genes encoding proteins from viruses, enabling controlled production of these antigens for immunology and infection research. When introduced into suitable host cells, the plasmid uses the cell’s transcription and translation machinery to express the viral protein, which can then be analyzed for antigen structure, immune recognition, or functional activity. Researchers use these constructs to study antibody and T-cell responses, evaluate diagnostic assays, investigate host-pathogen interactions, and support vaccine development. Their flexible design also enables comparison of viral variants and targeted modification of antigen sequences under defined laboratory conditions.

The video demonstrates an assay using human induced pluripotent stem cell (iPSC)-derived CD8+ T cells to inhibit hepatitis B virus (HBV) replication in mice. These T cells, engineered with HBV-specific receptors, are transferred into transgenic mice. Replication-competent HBV genome plasmids are then injected directly into the liver, enabling viral DNA uptake. Once viral antigens are expressed and presented by hybrid MHC-I molecules, the engineered T cells recognize and bind to these complexes.

A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules.
