Pmn Neutrophils

PMN neutrophils, also called polymorphonuclear neutrophils or neutrophil granulocytes, are the most abundant circulating white blood cells and a rapid first-line defense against infection. In response to inflammatory signals, they undergo chemotaxis toward affected tissues, where they recognize and engulf microbes through phagocytosis, release antimicrobial granules and reactive oxygen species, and can form extracellular traps that immobilize pathogens. In immunology and infection research, PMN neutrophils are studied to understand host defense, acute inflammation, tissue injury, and immune dysregulation. Their abundance, short lifespan, and functional responses also make them important targets in diagnostic assays and studies of infectious and inflammatory disease.

Quantitative Assessment of Human Neutrophil Migration Across a Cultured Bladder Epithelium

We developed an in vitro model that mimics an important component of the acute inflammatory response during infection of the bladder with uropathogenic Escherichia coli. The transuroepithelial neutrophil migration assay enables quantitative assessment of human neutrophil migration across bladder epithelia, cultured on permeable supports, in response to bacterial infection or chemoattractant substances.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
