Viral Protease Assay

A viral protease assay is a laboratory method that measures the activity of proteases encoded by viruses, enzymes that cleave specific protein substrates during infection and often support viral maturation. In a typical assay, purified protease or a viral sample is incubated with a peptide substrate containing a recognition sequence; cleavage produces a measurable fluorescent, colorimetric, or other signal proportional to enzyme activity. These assays help characterize viral replication mechanisms, compare protease variants, and evaluate candidate antiviral inhibitors. In immunology and infection research, they support studies of pathogen biology, drug potency, resistance, and host-pathogen interactions.

The Protease Fluorescent Detection Kit is designed for the measurement of protease activity using fluorometry. It is also suitable for detection of trace amounts of protease contamination. The method is based on the proteolytic hydroysis of a proprietary formulation of a FITC-labeled casein substrate.

This video demonstrates an assay to screen for the proteolytic activity of proteases using fluorogenic peptides. The protease recognizes its cleavage site on the peptide, cleaving it and separating the quencher from the fluorophore, enabling its fluorescence emission. The fluorescence signal is detected and analyzed to check for the cleavage efficiency of different peptide variants.

The Virochip is a pan-viral microarray designed to simultaneously detect all known viruses as well as novel viruses on the basis of conserved sequence homology. Here we demonstrate how to run a Virochip assay to analyze clinical samples for the presence of both known and unknown viruses.
