Viral Rna Sequencing

Viral RNA sequencing is a molecular technique used to determine the genetic sequence of RNA viruses, providing insight into viral identity, evolution, and interactions with the host immune system. Because RNA is unstable and cannot be read directly by most sequencing platforms, researchers first convert it into complementary DNA through reverse transcription, amplify target material, and sequence the resulting library using high-throughput methods. In immunology and infection research, these data help identify emerging variants, track transmission, measure viral diversity, and characterize mutations linked to replication or immune escape. The approach supports outbreak surveillance, diagnostic development, vaccine design, and studies of virus-host dynamics.

Detection of Viral RNA by Fluorescence in situ Hybridization (FISH)

A fluorescence in situ hybridization (FISH) method was developed to visually detect viral genomic RNA using fluorescence microscopy. A probe is made with specificity to the viral RNA that can then be identified using a combination of hybridization and immunofluorescence techniques. This technique offers the advantage of identifying the localization of the viral RNA or DNA at steady-state, providing information on the control of intracellular virus trafficking events.
