Conjunctival Melanoma

Conjunctival melanoma is a rare malignant tumor arising from melanocytes in the conjunctiva, the transparent membrane covering the eye and lining the inner eyelids. It develops when these pigment-producing cells undergo genetic changes that promote uncontrolled growth, producing a pigmented or nonpigmented lesion that may spread across the ocular surface or into deeper tissues. Clinical evaluation uses slit-lamp examination, documentation of lesion features, and, when indicated, biopsy with histopathologic and molecular assessment. Early recognition supports complete local treatment, such as excision with adjunctive therapy, while continued surveillance helps detect recurrence, regional spread, or distant metastasis and guides ophthalmic oncology care.

A rapid way to screen for melanoma modifiers using a zebrafish autochthonous tumor model is presented. It takes advantage of the miniCoopR vector which allows for expression of candidate melanoma genes in melanocytes. A method to obtain melanoma-free survival curves, an invasion assay, a protocol for antibody staining of scale melanocytes and a melanoma transplantation assay are described.
