Epididymal Adipose Tissue

Epididymal adipose tissue is a visceral fat depot located around the epididymis, a structure that supports sperm maturation, and it provides a useful model for studying adipose biology and metabolic disease. Its adipocytes store and mobilize triglycerides in response to insulin and energy demand while secreting adipokines, free fatty acids, and inflammatory mediators that can influence nearby tissues and whole-body metabolism. In medicine, researchers analyze this depot in animal studies of obesity, insulin resistance, inflammation, reproductive function, and drug effects, helping connect changes in fat distribution with metabolic and reproductive outcomes.

We present a protocol to isolate adipose tissue-derived microvascular fragments that represent promising vascularization units. They can be rapidly isolated, do not require in vitro processing and, thus, may be used for one-step prevascularization in different fields of tissue engineering.

Adipose tissue (AT) is a site of intense immune cell activation and interaction. Almost all cells of the immune system are present in AT and their ratios are altered by obesity. Proper isolation, quantification, and characterization of AT immune cell populations are critical for understanding their role in immunometabolic disease.
