Follicular Keratotic Plugs

Follicular keratotic plugs are compact accumulations of keratin and shed skin cells that obstruct the openings of hair follicles, producing rough, raised, or darkened papules on the skin. They form when abnormal keratinization, or retention of keratin within the follicular canal, prevents normal shedding and may trap sebum and microorganisms. These plugs are characteristic of conditions such as keratosis pilaris and comedonal acne, although their appearance and associated inflammation vary. Recognizing their distribution and clinical features helps clinicians distinguish common follicular disorders, select appropriate treatments that normalize keratin shedding, and assess whether additional evaluation is needed.

Bilayer Microfluidic Device for Combinatorial Plug Production

The fabrication of a polydimethylsiloxane (PDMS)-based bilayer device for the production of combinatorial libraries in water-in-oil emulsions (plugs) is presented here. The necessary hardware and software required to automate plug production are detailed in the protocol, and the production of a quantitative library of fluorescent plugs is also demonstrated.
