Joint Trauma Model

A joint trauma model is an experimental system that reproduces injury to a joint so researchers can study tissue damage, repair, and the progression of post-traumatic disease. Depending on the research question, trauma is induced through controlled mechanical impact or procedures that destabilize the joint, triggering cartilage disruption, inflammation, synovial responses, and changes in bone and ligament tissues. These models help clarify how an initial injury alters joint biology over time and provide platforms for evaluating imaging methods, biomarkers, rehabilitation strategies, and potential therapies. In medicine, joint trauma models support research into osteoarthritis prevention and improved treatment after musculoskeletal injury.

Acute brain trauma is a severe injury that has no adequate treatment to date. Multiphoton microscopy allows studying longitudinally the process of acute brain trauma development and probing therapeutical strategies in rodents. Two models of acute brain trauma studied with in vivo two-photon imaging of brain are demonstrated in this protocol.

We provide detailed methods for generating four types of tissues from human mesenchymal stem cells, which are used to recapitulate the cartilage, bone, fat pad, and synovium in the human knee joint. These four tissues are integrated into a customized bioreactor and connected through microfluidics, thus generating a knee joint-on-a-chip.
