Long-term Preservation

Long-term preservation is the maintenance of biological materials, medicines, or clinical specimens in a stable condition for extended periods, supporting future diagnosis, treatment, and research. It works by limiting chemical, enzymatic, and microbial degradation through controlled conditions such as low temperature, reduced moisture, protected packaging, and carefully selected storage media; cryopreservation may also use freezing protocols and cryoprotective agents to reduce cellular damage. In medicine, these strategies help preserve blood products, vaccines, tissues, cells, pharmaceuticals, and patient samples for biobanking, transplantation, quality testing, and longitudinal studies. Reliable preservation improves material availability, reproducibility, and continuity of care.

This paper presents a complete methodology to prepare and preserve in vitro acute hippocampal slices from adult mice. This protocol allows the recording of very stable long-lasting long-term potentiation (LTP) for more than 8 hours with a success rate of 95%.

Technological advances have led to optimizing the conditions of preservation, along with a more accurate assessment of the donor heart before transplantation. In this video, we describe the procedure for vitrification of harvested rabbit embryos at desired development stage using ethylene glycol and dimethyl sulfoxide as cryoprotectants and cryogenic straw as the carrier system.
