Lymphatic Malformation

Lymphatic malformation is a congenital disorder in which abnormal lymphatic vessels or spaces form and collect fluid, often producing a soft, compressible mass in the neck, face, or other tissues. These lesions arise from disrupted development of the lymphatic system, creating poorly connected channels that can enlarge with infection, bleeding, or increased lymph flow. Clinical evaluation and imaging, particularly ultrasound or magnetic resonance imaging, help define their extent and relationship to nearby structures. Treatment may include observation, sclerotherapy, surgical removal, or combinations of these approaches, with management guided by symptoms, location, growth, and effects on breathing, swallowing, or function.

The goal of this protocol is to isolate lymphatic endothelial cells lining human lymphatic malformation cyst-like vessels and foreskins using fluorescence-activated cell sorting (FACS). Subsequent cell culturing and expansion of these cells permits a new level of experimental sophistication for genetic, proteomic, functional and cell differentiation studies.

In this video, a genetically modified mouse pup carrying the Ccm2 gene flanked by loxP sites and Cre recombinase fused to an estrogen receptor is used to induce cerebral cavernous malformations (CCMs).
