Manual Double Centrifugation

Manual double centrifugation is a laboratory sperm-preparation technique that uses two sequential centrifugation steps to separate spermatozoa from seminal plasma, cellular debris, and other unwanted components. During the process, the sample is centrifuged to concentrate sperm into a pellet, the supernatant is removed, and the pellet is resuspended in a suitable culture medium before a second centrifugation and final resuspension. In medicine, this method supports sperm washing for assisted reproductive procedures by producing a more concentrated preparation for analysis or clinical use. Careful handling of centrifugation conditions is important because excessive mechanical stress can affect sperm quality and recovery.

We present a simple and efficient protocol for the generation of human macrophages. Buffy coats are processed by double density gradient centrifugation and isolated monocytes are then differentiated to macrophages in Teflon-coated cell culture bags. This maximizes macrophage yields and facilitates cell harvesting for subsequent experiments.
