Neonatal Asphyxia Model

A neonatal asphyxia model is an experimental system that reproduces the oxygen and blood-flow deficiencies associated with perinatal asphyxia, a major cause of newborn brain injury. By inducing controlled hypoxia-ischemia, often followed by restoration of oxygen and circulation, the model mimics the cellular stress, energy failure, inflammation, and neurological damage that can follow interrupted birth-related oxygen supply. Researchers use these models to investigate mechanisms of hypoxic-ischemic injury, evaluate biomarkers, and test interventions such as therapeutic hypothermia and neuroprotective treatments. The findings support improved understanding of neonatal encephalopathy and the development of strategies to reduce long-term disability.

Large animal models have good translational values in the examination of physiology and pharmacology of neonatal asphyxia. Using newborn piglets, we develop an experimental protocol to simulate neonatal asphyxia which has advantages of studying the systemic and regional hemodynamics, oxygen transport with biochemical and pathologic pathways and correlations.

Source: Seman, B. G., et al. A Neonatal Imaging Model of Gram-Negative Bacterial Sepsis. J. Vis. Exp. (2020)The video demonstrates a neonatal mouse model for studying systemic infection using encapsulated bioluminescent bacteria. Neonatal pups are injected with high- or low-dose bacterial suspensions and returned to their mother for postnatal care. The encapsulated bacteria evade early immune detection, spread systemically, and are visualized over time using luminescence imaging. This model...
