Peritoneal Macrophage Isolation

Peritoneal macrophage isolation is a laboratory technique used to obtain macrophages from the peritoneal cavity for studying innate immunity, inflammation, and disease processes. The procedure typically uses peritoneal lavage to recover resident immune cells, followed by centrifugation and cell culture conditions that support macrophage adherence and separation from nonadherent cells. Isolated macrophages can then be analyzed for morphology, viability, surface markers, phagocytosis, cytokine production, and responses to drugs or inflammatory stimuli. In medicine, this approach provides an accessible ex vivo model for investigating host defense, infection, tissue injury, and immune-mediated disorders while reducing the complexity of whole-animal experiments.
