Spontaneous Mutation Emergence

Spontaneous mutation emergence is the generation of genetic changes without deliberate exposure to an external mutagen, providing a fundamental source of biological variation relevant to medicine. These mutations can arise from DNA replication errors, endogenous chemical damage, or imperfect DNA repair, and they may alter a gene, regulatory sequence, or chromosome. Most have little detectable effect, but some influence cellular function, disease susceptibility, tumor evolution, or the response of microbes to treatment. Studying their formation and selection through genetic analysis and sequencing helps researchers interpret inherited disease, track cancer development, understand antimicrobial resistance, and improve strategies for diagnosis and therapy.

Spontaneous mutations arise infrequently during DNA replication due to errors in the process. A key factor behind these errors is tautomeric shifts in nitrogenous bases, where bases transition from keto to enol forms or amino to imino forms. This shift can alter base-pairing rules, leading to mutations. Additionally, reactive oxygen species (ROS) arising from aerobic metabolism can damage DNA, resulting in depurination (loss of a purine base) or depyrimidination (loss of a pyrimidine base).
