Vaginal Intraepithelial Neoplasia

Vaginal intraepithelial neoplasia (VaIN) is a precancerous condition in which abnormal squamous cells develop in the vaginal lining, making it important for identifying patients at risk of vaginal cancer. It most often reflects persistent infection with high-risk human papillomavirus (HPV), which can disrupt epithelial cell-cycle control and produce lesions graded by the extent of abnormal maturation and cellular atypia. Clinicians evaluate suspected VaIN with pelvic examination, cytology, HPV testing, colposcopy, and biopsy, then tailor management to lesion grade, size, symptoms, and patient factors. Observation may suit selected low-grade lesions, whereas laser ablation, excision, or topical therapy can treat higher-risk or persistent disease; surveillance is essential because recurrence and progression can occur.

Here, we performed a systematic evaluation of patients diagnosed with gastric low-grade intraepithelial neoplasia by previous endoscopic biopsy and obtained a pathological diagnosis by complete resection of the lesion by endoscopic submucosal dissection (ESD), analyzing factors that potentially increase the risk of pathological escalation.

Intravital Visualization of Gamma Delta Intraepithelial Lymphocytes

2025

This video demonstrates the visualization of γδ intraepithelial lymphocytes (IELs) in transgenic mice using confocal microscopy. The surgical procedure involves creating a perforation line and making a longitudinal incision to expose the intestinal mucosa and villi. The transgenic mice express reporter proteins exclusively in γδ IELs, allowing their tracking. Intravital imaging captures the dynamic movement of green fluorescence-labeled γδ IELs along the intestinal epithelial cell's basement...

Protocols for Vaginal Inoculation and Sample Collection in the Experimental Mouse Model of Candida vaginitis

Key techniques to be used in the evaluation of Candida vaginitis in an experimental animal model are described. The methods will allow rapid collection of vaginal specimens and lymphocytes from draining lumbar lymph nodes. These techniques could give rise to mouse models of other diseases in the female lower genital tract.

Modeling Murine Vaginal Colonization by Anaerobic Bacteria

Source: Morrill, S. R., et al. Models of Murine Vaginal Colonization by Anaerobically Grown Bacteria. J. Vis. Exp. (2022)This video demonstrates a protocol for modeling vaginal colonization in mice using antibiotic-resistant anaerobic bacteria. It begins with hormonal treatment to increase epithelial susceptibility, followed by vaginal inoculation and co-inoculation under anaerobic conditions. The procedure concludes with post-inoculation sampling and culture to evaluate bacterial colonization...

The purpose of this protocol is to imitate human group B Streptococcus (GBS) vaginal colonization in a murine model. This method may be used to investigate host immune responses and bacterial factors contributing to GBS vaginal persistence, as well as to test therapeutic strategies.
