Antibody-mediated Uptake

Antibody-mediated uptake is the internalization of antibody-bound molecules by cells, a process that can determine how biologics enter and move through neural tissue. It begins when an antibody recognizes a cell-surface antigen or receptor; binding can promote receptor clustering and endocytosis, after which vesicles traffic the antibody and its cargo through endosomal compartments or toward transcytosis across cellular barriers. In neuroscience, this process supports targeted delivery, cellular labeling, and investigation of blood-brain barrier transport, neuronal connectivity, and disease-associated proteins. Understanding uptake efficiency and intracellular routing helps improve antibody-based imaging and therapeutic strategies.

2025

This video demonstrates an assay measuring the antibody-mediated activation of T cell effector functions. Adding an influenza virus hemagglutinin (HA)-specific monoclonal antibody to transfected mammalian cells expressing HA results in the formation of immune complexes with HA. Upon introducing engineered T cells expressing Fc receptors and a luciferase reporter, the T cells are activated by the antibody-HA complexes, and this activation is detected by adding a luciferase substrate and...

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.

An Antibody Uptake Assay for Imaging Notch/DeltaD Signaling in Zebrafish Radial Glia Progenitors

This video demonstrates an antibody uptake assay to image Notch/DeltaD signaling in zebrafish radial glia progenitors. It involves mounting agarose-embedded dechorionated zebrafish embryos, injecting a dye solution targeting DeltaD ligands on RGPs, and imaging under a fluorescence microscope for asymmetric cell division confirming Notch/DeltaD signaling activation.
