Closed-head Injury Model

The closed-head injury model is an experimental approach that reproduces traumatic brain injury by applying mechanical force to the intact skull, enabling researchers to study brain damage without surgically exposing the brain. Impact-driven deformation transfers energy through the skull, producing tissue strain, axonal disruption, vascular changes, neuroinflammation, and altered neuronal function that can develop over time. In neuroscience, this model helps characterize acute and long-term consequences of head trauma, including cognitive, motor, and behavioral deficits. It also supports investigation of injury mechanisms, biomarkers, recovery processes, and potential treatments under controlled experimental conditions.

Source: Logsdon, A. F., et al. Low-intensity Blast Wave Model for Preclinical Assessment of Closed-head Mild Traumatic Brain Injury in Rodents. J. Vis. Exp. (2020)This video demonstrates the induction of closed-head mild traumatic brain injury in a mouse model using a pressurized gas shock tube setup. The generated pressure wave ruptures the polyester membrane, delivering a shockwave to the mouse’s head that results in rapid brain movement within the skull, causing a mild traumatic brain injury.
